Kim, Ki-Han (Wayne State University, Detroit, Mich.). Properties and distribution of intracellular putrescine in a Pseudomonas. J. Bacteriol. 91:193-197. 1966.-A Pseudomonas species which contains putrescine as the only intracellular polyamine was used to study the distribution of putrescine in the cells and the changes in putrescine content upon nitrogen or carbon and nitrogen starvation. In the cell-free extract, approximately 80 to 90% of the putrescine was found in the soluble fraction, and the rest was found in the ribosomal fraction; 50% of the putrescine could be removed from the cells by nitrogen starvation. Putrescine content in the ribosomes prepared from nitrogen-starved cells was about one-half of that in the unstarved cells. Putrescine was found in both 30S and 50S ribosomal particles. In the presence of 10(-3)m Mg(++), the ribosomal particles did not exchange bound putrescine for free putrescine, but did incorporate free spermine from the medium. Cells grown on glucose-NH(3) medium contained large amounts of acetyl putrescine. Cells grown on putrescine contained negligible amounts of acetyl putrescine, but readily formed acetyl putrescine when subjected to starvation.

Spermidine synthesis by Pseudomonas sp. strain Kim, previously reported to lack this polyamine.

Pseudomonas sp. strain Kim has previously been reported to be the only known naturally occurring organism lacking spermidine. We now show that it synthesizes this polyamine. The apparent lack of intracellular levels of spermidine results from an efficient conversion of spermidine to putrescine and hydroxyputrescine.

Difluoromethylornithine irreversibly inactivates ornithine decarboxylase of Pseudomonas aeruginosa, but does not inhibit the enzymes of Escherichia coli.

DL-alpha-Difluoromethylornithine, an enzyme-activated irreversible inhibitor of eukaryotic ornithine decarboxylase and consequently of putrescine biosynthesis, inhibited ornithine decarboxylase in enzyme extracts from Pseudomonas aeruginosa in a time-dependent manner t1/2 1 min, and also effectively blocked the enzyme activity in situ in the cell. Difluoromethylornithine, however, had no effect...

The Effect of Putrescine and Salicylic Acid on Physiological Characteristics and Antioxidant in Stevia Rebaudiana B. Under Salinity Stress

In order to investigate the effect of putrescin and salicylic acid on some physiological and antioxidant properties of stevia under salinity stress, a factorial experiment was conducted in a completely randomized design with three replications. In this experiment, samples were exposed to different concentrations of putrescine (0, 0.5 and 1 mM), salicylic acid (0 and 0.5 mM) and salinity (0, 75 ...

Bacteriol. 86:320-323. 1963.-A mutant was isolated from Escherchia coli B which can grow on putrescine as the sole source of carbon and nitrogen. This mutant contains a constitutive putrescine alpha-ketoglutarate transaminase and can be induced for the enzymes ...
